The ABSciex 4000QTrap is a hybrid triple quadrupole-linear ion trap mass spectrometer with a variety of available scan modes. ESI, APCI, and APPI ion sources are available. We find that the ESI and APPI sources complement each other well; the APPI source better ionizes several lipid classes that are not ionized as well by ESI. The system is interfaced with a Dionex UltiMate 3000 liquid chromatograph with ternary gradient capabilities, temperature-controlled autosampler, thermostatted column oven, and PDA detector.
The Apex-ultra 70 hybrid fourier transform mass spectrometer (FTMS) - Bruker Daltonics - is equipped with a 7.0 T actively shielded, high-stability magnet, and an Apollo II Dual ESI/MALDI source to provides simultaneous operation in both modes. The ESI source has an ion funnel and a spray chamber with a grounded off-axis sprayer. The MALDI source has computer controlled X-Y stage for scout-385 MTP target plates. Additional instrument capabilities are HPLC and nanospray experiments.
The ThermoFisher LTQ operated in nano LC mode is routinely used in tandem mass spectrometry experiments to identify peptides present at sub-femtomole levels. In practice, we can identify proteins in gel bands that are present at the lower levels detectable by silver staining (~1 ng). The instrument is also operated at higher flow rates for other applications such as H-D exchange. This instrument is generally operated by facility staff but some highly experienced users are granted direct access.
The LTQ-Orbitrap-Velos, representing the latest generation of high resolution mass spectrometers, possesses different ion fragmentation capabilities including CID, ETD, and HCD. The instrument is particularly useful for applications that require special fragmentation mechanisms, high resolution (100,000 ppm), high sensitivity, and/or rapid duty cycle.
